For what it's worth, I've been having the same problem for several releases of MacPerl now (for at least six months.) Rebuilding the desktop does NOT seem to help. The only thing that works is to manually open the MacPerl app, and then droplets work OK (or Alpha can find MacPerl, etc.) This is on a 7200, running OS8, but previously on 7.6. I never tried tracking down extension conflicts, though. (Sorry this isn't an answer, but at least you're not crazy...)
